I would think it would be no problem. At the worst, you would have to pull the wheels off for clearance.

I was able to pull mine off while the boat was still on but I think it depends on the how it is secured to the trailer (and mine was a single axle). If you at least remove the guide posts and then any of the screws that hold the fender down, you might be able to slip it out.
The only thing I can think of is accessing the screws that are under the boat hull. If you can access and remove those, you might have a shot at it. If I had all the screws removed, I could do it...94' 205 single axle trailer.

Mine is a single axle and both fenders had damage so I had to pull both. Problem for me was the upright guide poles. It was locked tight in the cup and had to heat the thing big time to get it out--didn't need the screws it was so locked in. Once the poles are out it isn't any problem at all. If you have an issue with the guide poles and have no way to heat them you can squeeze the fender out but you may damage the inside corner when you twist them around to clear so you can slide them up the poles. Still need to get the pole out so you don't damge after you fix it.
Good luck--no problem to removed with boat off trailer!

Yup, I masked the entire area around the repairs... (a hole on the top of the fender, a crack that was 36" long in the front, and a corner that was previously fixed with bondo and some plastic from a sign? - all from the Previous Owner.) And in my garage I have an exhaust fan that I run non-stop with a dehumidifier (for the racecar and all the associated chemicals/gas) with heat and cooling to be able to use the garage all winter
Interestingly enough, I was able to buy Duplicolor white primer and Duplicolor Toyota white and blend in the repairs with the existing paint by feathering it in, wet sanding, and polishing... I did it over the winter and didnt have room to remove the fender. There is some thread somewhere that shows the progress in a few pictures (maybe in the what did you do to your MC today thread). As of today with a few more wet sands and polishing (no new pics yet); you cant even tell! :)
IF I was smarter (Hind-sight), I would have taken the fender offoff before I put it in the garage for winter....and sprayed the whole thing.

I should add it was an extra PIA because I ground out the cracks, holes, and cut out the junk and glassed it back in from the top and the bottom (upside down!! - ugh)

Mine were busted pretty bad but turned out well--also got decals redone by local shop except for one I had to cut out by hand.

If you pull all bolts and guide posts and wont just slide right out, you might pull wheel also, and then its a breeze!
